Estrogen receptor (ER) alpha belongs to the nuclear receptor (NR) superfamily of structurally related ligand-inducible transcription factors. The classical mechanism of activation of ERs is dependent on the ligand binding to the receptor, which causes receptor dimerization and then binding to estrogen response elements (EREs) located within the promoters of estrogen-responsive genes. Inappropriate activation of NRs such as ER is associated with numerous diseases. As such, these receptors have proven to be promising drug targets and today there are a number of therapeutic agents that directly influence NR protein activity.
