Contents: 1 mg (Catalog No. 15030) or 0.1 mg (Catalog No. 15050) of Chromeo™ 505 conjugated Goat anti-Mouse IgG (H+L). The antibody concentration is 2 mg/ml in 0.01 M potassium phosphate, 0.15 M sodium chloride pH 7.4, containing 2 mM sodium azide.

Specificity: This antibody was purified by immunoaffinity chromatography. It reacts with whole molecular mouse IgG and the light chains of other mouse immunoglobulins. No cross-reactivity with non-immunoglobulin serum proteins was observed.

Chromeo 505 can be excited in a range from 480-515 nm, which enables excitation by a green laser. Even if the emission spectrum is shifted towards longer wavelength by some nm, filters designed for fluorescein can be used with Chromeo 505 conjugated antibodies. In contrast to fluorescein, Chromeo 505 exhibits a large tolerance to a broad pH range and high photostability. The Chromeo 505 conjugated antibodies have absorption and emission maxima of approximately 514 and 530 nm, which are compatible with common excitation sources and filter sets.

Chromeo 505 Goat anti-Mouse IgG is recommended by Leica Microsystems for Super-Resolution Fluorescence Microscopy using Leica's STimulated Emission Depletion (STED) microscopes. It has also been tested and verified by Leica for use in Ground State Depletion with Individual Molecule return (GSDIM) using the SR GSD microscope. STED enables resolution of 50-70 nm, while GSDIM can mathematically re-construct images with resolution as low as 20 nm.

HeLa cells were stained with alpha Tubulin mouse mAb (Clone 5-B-1-2; Cat. No. 39527) and Chromeo 505 Goat anti-Mouse IgG.

Multi-color staining of Phospho H2AX and tubulin using Chromeo 488 and Chromeo 494 secondary antibodies in etoposide-treated HeLa cells.
HeLa cells were treated with 100 µM etoposide for 6 hours prior to fixation with methanol. The histone-variant H2AX was stained with Histone H2AX phospho Ser139 rabbit pAb (Cat. No. 39117) and Chromeo 642 Goat anti-Rabbit IgG (Cat. No. 15044), while tubulin was visualized using alpha Tubulin mouse mAb (Clone 5-B-1-2, Cat. No. 39527) and Chromeo 505 Goat anti-Mouse IgG. The nuclei were counter-stained with DAPI and then the three separate images were merged.
