Recombinant CK2α2 is suitable for kinase assays. The molecular weight of the protein is ~84.297 kDa. The activity of the protein is ~ 1,210,000 units/mg with 1 unit defined as the amount of enzyme that will catalyze the transfer of 1 pmol phosphate to synthetic peptide substrate RRRDDDSDDD per minute at 37°C. Recommended Kinase Reaction Buffer: 20 mM Tris-HCL, 25 mM β-glycerophosphate, 5 mM EGTA, and 1 mM DTT, pH 7.5. Kinase activity may vary depending on the substrate and reaction conditions used.

Recombinant human CK2α2 was expressed in E. coli as a N-MBP-fusion protein. The accession number is NM_001896. The alpha subunit is the catalytic subunit of serine/threonine protein kinase CK2 and is constitutively active. This protein suitable for labeling CK2α substrates. This protein has shown to be extremely salt sensitive and highest activity is obtained without salt. Purification by samylose affinity chromatography to > 95% by SDS-PAGE.
