15 µg of Recombinant Human Active JNK2α1 protein in 50 mM Tris-HCl (pH 7.5), 0.15 M NaCl, 0.27 M sucrose, 10 mM β-mercaptoethanol, 0.1% Triton X-100 and I mM EGTA, carrier-free. Protein is supplied at 0.31 µg/µl
Application Notes:
Recombinant human active JNK2α1 (amino acids 1-382) is produced in
E. coli with an amino terminal polyhistidine tag and purified via sequential chromatography to >95%. Recombinant JNK2α is capable of autophosphorylation and also phosphorylates c-Jun at serine 63 and serine 73. JNK2α. is shorter than JNK2α2 and contains a five nucleotide insertion in the C terminal region which causes a shift in the reading frame and subsequent premature termination.
Protein Usage:
Recombinant active JNK2α1 is suitable for kinase assays and Western blot. The molecular weight of the protein is ~45 kDa. The activity of the protein is ~125 units/mg and one unit of JNK2α1 activity is equal to 1 nanomole of phosphate transferred to recombinant ATF-2 fusion protein per minute at 30°C with a final ATP concentration of 100 µM. Kinase activity may vary depending on the substrate and reaction conditions used.
NOTE: The presence of Poly [d(I-C)] in buffers may affect protein functionality and should be avoided.
Storage:
Some proteins are supplied lyophilized and some are supplied in solution. Lyophilized proteins can be stored at 4°C, preferably desiccated. Recombinant proteins in solution are temperature sensitive and must be stored at -80°C to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
