Recombinant Set8 D338A was expressed as the human protein missing amino acids 13-17 (accession number NP_065115.3) with an N-terminal poly-histidine tag. The recombinant Set8 D338A protein contains a substitution of aspartic acid to alanine at amino acid 338. Set8 D338A is unable to methylate histone H4.
