for U-937 nuclear extract (IFNa treated)Background
U-937 nuclear extract (IFNα treated) was prepared from cell cultures of the human histiotic lymphoma U-937 cell line. The U-937 cell line was derived from malignant cells of a pleural effusion of a 37-year old male patient. These cells are of myeloid lineage and differentiate into a mature macrophage phenotype in response to various stimuli. U-937 cells produce and secrete large quantities of cytokines and chemokines either spontaneously or in response to treatment with soluble stimuli, such as TNFα. Furthermore, IFNβ and IFNϒ stimulation has been shown to induce the expression of chemokine receptors on the cell surface of U-937 cells and results in increased production of interleukins. Because U-937 cells are one of only a few human cell lines with characteristic monocytic properties, they are commonly used in research as in vitro models for monocyte/macrophage differentiation.
In addition to increasing the expression of chemokine receptors, treatment of U-937 cells with the IFNα induces cell cycle arrest and differentiation of U-937. IFNα treatment of U-937 cells also promotes apoptosis, causes alterations in viral replication efficiency, and induces the JAK/STAT signaling pathway.
for U-937 nuclear extract (IFNa treated)Application Notes
U-937 extract is recommended for studies of monocyte/macrophage differentiation. U-937 nuclear extract (IFNα treated) is specifically recommended for studies related to 1) chemokine and cytokine production and secretion, 2) proliferation and apoptosis, 3) viral replication (ie HIV), and 4) the JAK/STAT signaling pathway.

for U-937 nuclear extract (IFNa treated)Extract Composition
U-937 nuclear extract (IFNα treated) is supplied in Dilution Buffer (20 mM Hepes (pH 7.9), 100 mM KCl, 1 mM MgCl2, 20% glycerol, 0.5 mM PMSF and 0.5 mM DTT). Cells were cultured in medium supplemented with 5000 U/ml interferon alpha for one hour at 37°C immediately prior to harvesting.
for U-937 nuclear extract (IFNa treated)Quality Control
Extracts have been quality control tested by Western blot and the Electrophoretic Mobility Shift Assay (EMSA).
for U-937 nuclear extract (IFNa treated)Storage
To ensure stability, extracts should be stored at -80°C.
We recommend aliquoting the extracts into single-use fractions and then storing them at -80°C. This eliminates repeated freeze/thaw cycles.
