Background for p53 antibody (pAb):

p53 is the most important tumor suppressor in the genome. It is responsive to numerous genotoxic stresses, which activates its transcription factor activity, in turn causing cell-cycle arrest by activating expression of p21 Cip/WAF. Mutant p53 that has lost its DNA-binding function interferes with the activity of native p53 and leads to oncogenic transformation. Alternatively, transformation may be caused by overexpression of Mdm2/Hdm2, a ubiquitin ligase specific for p53, which causes its destabilization. Inactivation of p53 is often coincident with hyperactivation of NFκB (NFκB p50 and NFκB p65), both of which serve to inhibit apoptosis.

Immunogen for p53 antibody (pAb):

This p53 antibody was raised against a synthetic peptide corresponding to amino acid residues 367-381 of human p53.

Application Notes for p53 antibody (pAb):

This p53 antibody has been validated for use in Western blotting (1:500 - 1:1,000 dilution) and immunohistochemistry (1:100 dilution). For optimal results, primary antibody incubations should be performed at 37°C. Individual optimization may be required.

Buffer:

PBS containing 0.2% gelatin, 0.01% thimersol and 0.1% sodium azide. Thimersol and sodium azide are highly toxic.

Positive Control:

Active Motif's 3T6 Swiss albino nuclear extract (Catalog No. 36002) can be used as a positive control for p53 antibody.

Detection of p53 by Western blot analysis. The presence of the transcription factor p53 in nuclear extracts derived from 3T6 Swiss albino cells is confirmed using p53 rabbit polyclonal antibody at a 1:500 dilution (lane 1). The signal corresponding to nuclear p53 is eliminated when the immunizing peptide is added to the reaction (lane 2).

Storage:

Antibodies in solution can be stored at -80°C for 2 years. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
