for MBD4 antibody (pAb)Background
Methylation of mammalian DNA has long been recognized to play a major role in a number of cellular functions such as development and control of gene expression. It is generally associated with the repressive chromatin state. The complex series of events leading to this repressive state involve the coordinated regulation of DNA methyltransferases and two other groups of proteins called the Methyl-CpG binding proteins (MBD proteins) and the Kaiso family of proteins. The MBD family of proteins include MeCP2, MBD1, MBD2, MBD3 and MBD4. MBD4 (methyl-CpG binding domain protein 4) is a thymine glycosylase that recognizes the product of deamination at methyl-CpG sites, as a part of DNA repair system. MBD4 is able to bind the hemimethylated DNA or methyl-CpG TpG mismatches.
for MBD4 antibody (pAb)Immunogen
This antibody was raised against a mixture of synthetic peptides corresponding to amino acid residues 268-282 and 337-352 of human MBD4.
for MBD4 antibody (pAb)Application Notes
This MBD4 antibody has been validated for use in Western blotting (1 - 2 µg/ml dilution). For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all Blotto solutions may also reduce background. Individual optimization may be required.
for MBD4 antibody (pAb)Buffer
PBS containing 0.02% sodium azide. Sodium azide is highly toxic.
for MBD4 antibody (pAb)Positive Control
Active Motif's HL-60 nuclear extract (Catalog No. 36072) can be used as a positive control for MBD4 antibody.
MBD4 pAb tested by Western blot.
The analysis was performed using HL-60 whole-cell extract and MBD4 pAb at a 2 µg/ml dilution.
for MBD4 antibody (pAb)Storage
Antibodies in solution can be stored at -80°C for 2 years. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
for MBD4 antibody (pAb)Guarantee
