Cyk-4 is a member of a GTPase activating protein family that regulates the catalytic function of Rho and Ras small G-proteins. Cyk-4 regulates the initiation of cytokinesis through its interaction with a variety of microtubule organizing proteins. The activity of Cyk-4 is regulated by phophorylation of serine 170 by Polo-like Kinase 1.

Recombinant Cyk-4 fragment (100 ng) untreated (lane 1) and incubated with the kinase Plk-1 (lane 2) was probed with 39265 at 1:2,000.

Dot blot analysis was used to confirm the specificity of 39265 for phospho Ser170 Cyk-4. The modified and unmodified peptides to the immunogen were spotted onto PVDF and probed with 39265 at a dilution of 1:2,000. The amount of peptide (picomoles) spotted is 250,50,10 and 2 from top to bottom.
