Background for Pit-1 antibody (pAb):
Pit-1 (
POU1F1,
GHF-1,
growth hormone factor-1,
pituitary specific transcription factor 1) is a
pituitary-specific transcription factor responsible for pituitary development and hormone expression in mammals and is a member of the
POU family of transcription factors that regulate mammalian development. The POU family is so named because the first 3 members identified were
Pit-1 and Oct-1 (MIM 164175) of mammals, and Unc-86 of
C. elegans.
Pit-1 activates the production of growth hormone and
Prolactin. In addition,
Pit-1 acts as a
repressor of gene expression, which allows for the differentiation of specific cell types.
Pit-1 is expressed as two alternatively spliced products, designated Pit-1α and Pit-1β, that have molecular masses of 33 and 31 kDa, and differ in their trans-activation ability. Mutations in the
Pit-1 gene is believed to result in
combined pituitary hormone deficiency (
CPHD) for growth hormone, Prolactin and thyroid stimulating hormone.
Immunogen for Pit-1 antibody (pAb):
This antibody was raised against a synthetic peptide corresponding to amino acid residues 107-120 of human Pit-1.
Application Notes for Pit-1 antibody (pAb):
This Pit-1 antibody has been validated for use in electrophoretic mobility shift assays (EMSA). Sufficient reagent to perform 17 supershift reactions is provided. We recommend adding 2 µl of antibody to the reaction mixture when performing supershift experiments. We also recommend the use of Tris-glycine electrophoresis buffer. Individual optimization may be required. This antibody is not recommended for use in Western blotting.
Buffer:
PBS containing 0.2% gelatin, 0.01% thimersol and 0.1% sodium azide. Thimersol and sodium azide are highly toxic.
Positive Control:
Active Motif's
GH3 nuclear extract (Catalog No. 36008) can be used as a positive control for Pit-1 antibody.

Pit-1 pAb tested by supershift.
Detection of Pit-1 by EMSA analysis. Pit-1 pAb was used in a supershift assay (EMSA) with GH3 nuclear extract (lane 4). Lane 5 demonstrates a supershift competition in the presence of neutralizing peptide.
