MAD2L1 (aka HSMAD2, MAD2, MAD2-like 1) is the human homolog of the budding yeast MAD2, or mitotic arrest deficient 2 gene. MAD2L1 is a component of the mitotic spindle assembly checkpoint, which monitors the attachment of the mitotic spindle to the kinetochores of segregating chromosomes and delays the onset of anaphase until attachment is complete and the chromosomes are properly aligned on the metaphase plate. MAD2L1 associates only with unattached kinetochores in prometaphase, and prevents cell cycle progression by associating with the Anaphase Promoting Complex (APC), specifically restricting the activity of the CDC20 protein.
