Background for CTCF antibody (mAb) CCCTC-binding factor:
The vertebrate insulator protein, CCCTC binding factor (CTCF), is a highly conserved, ubiquitously expressed zinc-finger DNA-binding protein that has widespread functions in gene regulation, chromatin organization and epigenetic imprinting. CTCF plays dual roles in cell growth regulatory processes and epigenetic functions. CTCF is involved in different aspects of gene regulation including promoter activation and repression, hormone-responsive gene silencing, methylation-dependent chromatin insulation, and genomic imprinting. CTCF can modulate higher-order chromatin structure by regulating long-distance interactions between proteins bound at discrete regulatory elements.Chromatin IP Assays:

This CTCF antibody (mAb) has been validated for use in Western blotting (0.1 - 1 µg/ml) and chromatin IP (0.2 - 2 µg per ChIP).The development of this antibody is described in Reference 1 (Kim, et al, 2007).
Buffer:
Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 31 mM glycine, 0.07 mM EDTA, 30% glycerol, and 0.035% sodium azide.Positive Control:

CTCF antibody (mAb) tested by Western blot.
Nuclear extract of HEK293 cells (20 µg per lane) probed with CTCF antibody (mAb) at a dilution of 0.25 µg/ml.
CTCF antibody (mAb) tested by ChIP.
Chromatin IP performed using the ChIP-IT™ Express Kit (Catalog No. 53008) and 25 µl of Ready-to-ChIP HeLa Chromatin (Catalog No. 53015) per ChIP using 2 µg of CTCF mAb or 2 µg of mouse IgG (IgG) as a negative control. Real time, quantitative PCR (RT-qPCR) was performed on DNA purified from each of the ChIP reactions using a primer pair specific for the Imprinting Control Region (ICR) of the IGF2/H19 locus. The data presented are expressed as a percentage of the starting material (ChIP/input).
