DNMT3L (DNA methyltransferase 3-like) is a nuclear protein with similarity to the DNMT family of proteins, but does not function as a DNA methyltransferase. It serves as an essential regulator of DNMT3A and DNMT3B, recruiting them to sites for deposition of de novo DNA methylation. DNMT3L is expressed in the presumptive gametes in mammalian embryos and is required for the establishment of genomic imprinting. DNMT3L has a PHD-like domain that allows binding to chromatin through interactions with the amino terminal tail of histone H3 that is devoid of lysine 4 methylation.
