for SREBP-2 antibody (mAb) (Clone IgG-1C6)Background

SREBP-1 and SREBP-2 (Sterol regulatory element-binding protein 1 and 2) are members of the basic-helix-loop-helix-leucine zipper (bHLH-ZIP) family of transcription factors and are involved in the regulation of cholesterol homeostasis. SREBP-1 and SREBP-2 are normally membrane bound at the ER and nuclear envelope, but in sterol-depleted cells, the membrane-bound precursors are cleaved to generate a soluble isoforms that translocate to the nucleus. These active forms, which include the bHLH-ZIP domain, activate transcription of genes involved in the uptake and synthesis of cholesterol.

This SREBP-2 antibody was raised against a recombinant protein corresponding to amino acids 833-1141 of human SREBP-2.

for SREBP-2 antibody (mAb) (Clone IgG-1C6)Application Notes

This SREBP-2 antibody has been validated for use in chromatin immunoprecipitation (ChIP, 1 - 2 µg per ChIP), immunoprecipitation (1 - 2 µg per IP) and Western blotting (0.5 - 2 µg/ml). This antibody recognizes the 125 kDa precursor and the processed 68 kDa nuclear form.

Bridging Antibody to Improve ChIP, MeDIP and IP

Some isotypes of mouse IgG antibodies do not bind well to protein G-conjugated agarose/magnetic beads, which are commonly used in ChIP, MeDIP and IP experiments. Capture efficiency can be improved by including an anti-mouse IgG bridging antibody because it binds with a strong affinity to both the protein G beads and the mouse IgG antibody. By increasing the affinity of the mouse primary antibody for the protein G bead, the Bridging Antibody for Mouse IgG can help improve the results of all ChIP, MeDIP and IP experiments that use SREBP-2 antibody (mAb).
