RANK (TNFRSF11A, CD265, PDB2, TRANCER) can interact with various TRAF family proteins (TRAF2, TRAF5), through which this receptor induces the activation of NFκB and MAPK8/JNK. RANK and its ligand are important regulators of the interaction between T cells and dendritic cells. RANK is also an essential mediator for osteoclast and lymph node development. Studies of the mouse counterpart suggest that this receptor directly mediates the osteoprotegerin (OPG) ligand (OPGL)-induced oteoclastogenesis in osteoclast precursor cells.

This RANK antibody has been validated for use in Western blotting (1 - 2 µg/ml dilution). For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all Blotto solutions may also reduce background. Individual optimization may be required.

RANK detection by Western blot. The analysis was performed using 293 cells transfected with RANK cDNA and RANK mAb at a 2 µg/ml dilution. A single protein band of approximate mol. wt. of 97 kDa was detected.
