Caspase-8 (CASP8 or apoptosis-related cysteine peptidase 8) is the most upstream protease of the activation cascade of caspases responsible for the TNFRSF6/FAS mediated and TNFRSF1A induced cell death. Binding to the adaptor molecule FADD recruits it to either receptor. Caspase-8 cleaves and activates Caspase-3, Caspase-4, Caspase-6, Caspase-7, Caspase-9 and Caspase-10.

This Caspase-8 antibody has been validated for use in Western blotting (1 - 2 µg/ml dilution), flow cytometry (0.1 µg) and immunohistochemistry (5 µg/ml dilution). For optimal results in Western blotting, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all Blotto solutions may also reduce background. Individual optimization may be required.

Caspase-8 detection by Western blot. The analysis was performed using Jurkat extracts with Caspase-8 mAb at 2 µg/ml (lane 1) and 0.5 µg/ml (lane 2) dilutions. The antibody only detects 55 kDa caspase-8 in staurosporine-treated (2 hours) Jurkat cells.
