D4-GDI (Rho GDP dissociation inhibitor beta or ARHGDIB) plays an important role in the activation of the superoxide (O2-)-generating NADPH oxidase of phagocytes. The mechanisms involving cleavage of D4-GDI (Rho GDP) with apoptosis are not presently known. However, activation of the Jun N-terminal kinase, a regulator of apoptosis, may be one of the mechanisms.

This D4-GDI antibody has been validated for use in Western blotting (1 - 2 µg/ml dilution), and reported to work in immunoprecipitation. For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all Blotto solutions may also reduce background. Individual optimization may be required.

Bridging Antibody to Improve ChIP, MeDIP and IP
Some isotypes of mouse IgG antibodies do not bind well to protein G-conjugated agarose/magnetic beads, which are commonly used in ChIP, MeDIP and IP experiments. Capture efficiency can be improved by including an anti-mouse IgG bridging antibody because it binds with a strong affinity to both the protein G beads and the mouse IgG antibody. By increasing the affinity of the mouse primary antibody for the protein G bead, the Bridging Antibody for Mouse IgG can help improve the results of all ChIP, MeDIP and IP experiments that use D4-GDI antibody (mAb).
D4-GDI mAb tested by Western blot.
Detection of D4-GDI by Western blot analysis. HL-60 cells were treated for different time periods with 10 ng/ml of Fas monoclonal antibody. Harvested cells were lysed, resolved by SDS-PAGE and transferred onto a PVDF membrane. The membrane was incubated with the D4-GDI mAb and immunoreactivity was detected by chemiluminescence. The data shows that the D4-GDI antibody specifically detects the 23 kDa cleaved form of D4-GDI in a time-sensitive manner.
