Background for p73DN antibody (mAb):
p73ΔN is a member of the
p53 protein family which includes p53 as well as
p63. Whereas p53 transactivates the retinoblastoma gene, p63 and
p73 induce the cyclin-dependent kinase inhibitor p57 to maintain
Rb in an active, hypophosphorylated state.
p73ΔN inhibits the functions of the p53 family in differentiation control, prevents myogenic differentiation, and enables cooperating oncogenes to transform myoblasts to tumorigenicity.
p73ΔN is frequently overexpressed in rhabdomyosarcoma and is essential for tumor progression
in vivo. These findings establish differentiation control as a key tumor suppressor activity of the p53 family.
Immunogen for p73DN antibody (mAb):
This p73DN antibody was raised against a peptide corresponding to amino acid residues 2-13 of human p73ΔN.
Application Notes for p73DN antibody (mAb):
This p73DN antibody has been validated for use in Western blotting (1 - 2 µg/ml dilution). For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all Blotto solutions may also reduce background. Individual optimization may be required. This antibody does not cross react with full-length p73.
Buffer:
PBS containing 0.02% sodium azide. Sodium azide is highly toxic.
Positive Control:
Extracts from cells transfected with p73ΔN cDNA can be used as a positive control for p73ΔN antibody.
