Applications

Western Blot Validated Immunofluorescence Validated Immunocytochemistry Validated

Application Notes

Applications Validated by Active Motif:
ICC/IF: 2 - 10 µg/ml dilution
WB*: 1 - 2 µg/ml dilution

*Note: many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt / Sonication Protocol when preparing nuclear extracts for Western Blot.

This antibody is also available as an AbFlex® engineered recombinant antibody. For details on the corresponding AbFlex Recombinant Antibody, see Catalog No. 91251.

Immunogen

This antibody was raised against a peptide comprising amino acids 2-25 of mouse Nanog.

Buffer

Purified IgG in PBS with 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

Nanog antibody (mAb) (Clone 2E6E1) tested by Western blot.
Nanog antibody detection by Western blot. The analysis was performed using 30 µg of mouse ES nuclear extract and Nanog antibody at a 2 µg/ml dilution.

Background

Nanog (Nanog homeobox) is a transcriptional regulator involved in inner cell mass and embryonic stem (ES) cells proliferation and self-renewal. Imposes pluripotency on ES cells and prevents their differentiation towards extraembryonic endoderm and trophectoderm lineages. Blocks bone morphogenetic protein-induced mesoderm differentiation of ES cells by physically interacting with SMAD1 and interfering with the recruitment of coactivators to the active SMAD transcriptional complexes (By similarity). Acts as a transcriptional activator or repressor (By similarity). Binds optimally to the DNA consensus sequence 5'-TAAT[GT][GT]-3' or 5'-[CG][GA][CG]C[GC]ATTAN[GC]-3' (By similarity). When overexpressed, promotes cells to enter into S phase and proliferation (By similarity).
