The addition of 0.05% Tween 20 in the blocking buffer and primary antibody incubation buffer is recommended to aid in detection by Western blot. Individual optimization may be required.
Immunogen
This DNMT1 antibody was raised against a recombinant protein corresponding to full-length human DNMT3L.
Buffer
Purified IgG in PBS with 50% glycerol and 0.02% sodium azide. Sodium azide is highly toxic.

Nuclear extract (20 µg) of human testicular cells probed with DNMT3L antibody at a dilution of 1:500.
Background
DNMT3L (DNA methyltransferase 3-like) is a nuclear protein with similarity to the DNMT family of proteins, but it does not function as a DNA methyltransferase. It serves as an essential regulator of DNMT3A and DNMT3B, recruiting them to sites for deposition of de novo DNA methylation. DNMT3L is expressed in the presumptive gametes in mammalian embryos and is required for the establishment of genomic imprinting. DNMT3L has a PHD-like domain that allows binding to chromatin through interactions with the amino terminal tail of histone H3 that is devoid of lysine 4 methylation.
Storage
Some products may be shipped at room temperature. This will not affect their stability or performance. Avoid repeated freeze/thaw cycles by aliquoting items into single-use fractions for storage at -20°C for up to 2 years. Keep all reagents on ice when not in storage.
