For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all blocking solutions may also reduce background. Individual optimization may be required.
Immunogen
This Caspase-9 antibody (Pro-form) was raised against a peptide corresponding to amino acid residues 299 -318 of human Pro-Caspase-9.
Buffer
PBS containing 0.02% sodium azide. Sodium azide is highly toxic.
Caspase-9 pAb (Pro-form) tested by Western blot.
Caspase-9 detection by Western blot. The analysis was performed using 50 µg HeLa whole-cell extract and Caspase-9 pAb (Pro-form) at a 1:1,000 dilution.
Background
Caspase-9 (CASP9 or apoptosis-related cysteine peptidase 9) (Pro-form) is involved in the activation cascade of caspases responsible for apoptosis execution. Caspase-9 is processed by caspase APAF1, which is thought to be one of the earliest events in the caspase activation cascade. Caspase-9 is then involved in processing of Caspase-3.
Positive Control
Active Motif's HeLa whole-cell extract (Catalog No. 40050) can be used as a positive control for Caspase-9 antibody (Pro-form).
Storage
Some products may be shipped at room temperature. This will not affect their stability or performance. Store at 4°C for short term. Avoid repeated freeze/thaw cycles by aliquoting items into single-use fractions for storage at -20°C for up to 2 years. Keep all reagents on ice when not in storage.
