Applications Validated by Active Motif:
WB: 1 - 2 µg/ml dilution
For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all blocking solutions may also reduce background. Individual optimization may be required.
Immunogen
This Caspase-14 antibody was raised against a synthetic peptide corresponding to amino acid residues 2-18 of mouse Caspase-14.
Buffer
PBS containing 0.02% sodium azide. Sodium azide is highly toxic.
Caspase-14 mAb tested by Western blot.
Caspase-14 detection by Western blot. The analysis was performed using 10 µg of mouse spleen cell extract and Caspase-14 mAb at a 2 µg/ml dilution.
Background
Caspase-14 (CASP14 or apoptosis-related cysteine peptidase 14) is thought to be a non-apoptotic caspase involved in epidermal differentiation. Caspase-14 has been shown to be processed and activated by Caspase-8 and Caspase 10 in vitro, and also by TNF-Related Apoptosis-Inducing Ligand (TRAIL) in vivo.
Positive Control
Extracts from mouse spleen can be used as a positive control for Caspase-14 antibody.
Storage
Some products may be shipped at room temperature. This will not affect their stability or performance. Avoid repeated freeze/thaw cycles by aliquoting items into single-use fractions for storage at -20°C for up to 2 years. Keep all reagents on ice when not in storage.
