for SSRP1 antibody (pAb)Application Notes

Validated Applications:
WB: 1:500 - 1:2,000 dilution

The addition of 0.05% Tween 20 in the blocking buffer and primary antibody incubation buffer is recommended to aid in detection by Western blot. Individual optimization may be required.

for SSRP1 antibody (pAb)Background

SSRP1 (Structure-Specific Recognition Protein 1) is a component of the FACT complex, a general chromatin factor that acts to reorganize nucleosomes. SSRP1 and Supt16H form a stable heterodimer and this complex is involved in multiple processes that require DNA as a template such as mRNA elongation, DNA replication and DNA repair. During transcription elongation the FACT complex acts as a histone chaperone that both destabilizes and restores nucleosomal structure. It facilitates the passage of RNA polymerase II and transcription by promoting the dissociation of one histone H2A-H2B dimer from the nucleosome, then subsequently promotes the reestablishment of the nucleosome following the passage of RNA polymerase II. The FACT complex is probably also involved in phosphorylation of 'Ser-392' of p53/TP53 via its association with CK2 (casein kinase II). Binds specifically to double-stranded DNA and at low levels to DNA modified by the antitumor agent cisplatin. The HMG box DNA-binding domain mediates DNA-binding with both affinity and specificity for DNA damaged globally with cisplatin. May potentiate cisplatin-induced cell death by blocking replication and repair of modified DNA. Also acts as a transcriptional coactivator for p63/TP63.

for SSRP1 antibody (pAb)Positive Control

Active Motif's K-562 nuclear extract (Catalog No. 36015) can be used as a positive control for SSRP1 antibody.

for SSRP1 antibody (pAb)Storage

Some products may be shipped at room temperature. This will not affect their stability or performance. Upon receipt, unconjugated antibodies may be stored at -20°C for up to 2 years. Fluorophore- & enzyme-conjugated antibodies should be stored at 4°C. Fluorophore-conjugated antibodies should be protected from light. Keep reagents on ice when not in storage; to avoid repeated freeze/thaw cycles, we recommend aliquoting items that will be stored frozen into single-use fractions prior to freezing.
