LightSwitch Pathway Reporter Stable Cell Lines

LightSwitch Pathway Reporter Stable Cell Lines simplify the task of measuring the response of specific pathways to treatment with various compounds or growth conditions. Each cell line contains a human regulatory element that has been cloned into a LightSwitch reporter vector and validated for use in measuring the activity of a specific biological pathway. Because they can be used in a variety of different plate-based formats, the Pathway Reporter Stable Cell Lines are ideal for both primary and secondary high-throughput screening.

If we don't have the stable cell line you need, Custom Services are available for creating custom stable cell lines, as well as for conducting screening services. Active Motif also offers LightSwitch Validated Pathway Collections, which are sets of promoter reporter constructs that showed significant activation or repression in response to various pathway-specific induction conditions.

IMPORTANT:  Because all LightSwitch reporter constructs contain the optimized RenSP luciferase gene, you MUST use our LightSwitch Luciferase Assay Reagents to obtain optimal results. (Other luciferase assay reagents are not compatible with RenSP.)

LightSwitch Luciferase Pathway Reporter Stable Cell Line Overview

LightSwitch Pathway Reporter Stable Cell Lines make it easier to measure the response of a pathway to compounds and growth conditions because the regulatory element to be tested and the RenSP luciferase are already stably integrated into the cell's genome. Simply seed the cell line in multi-well plates, treat each well as desired, then perform high-throughput luciferase assays to measure the response of the promoter to the various treatments.

LightSwitch Luciferase Pathway Reporter Stable Cell Lines

The LightSwitch Pathway Reporter Stable Cell Lines are ideal tools for use in high-throughput screening. In contrast to transiently transfected reporter vectors, the reporter lines stably express the RenSP reporter gene regulated by an element that is responsive to a specific pathway. This eliminates the need to transfect the reporter construct and increases the length of time over which experiments can be conducted.

Applications

  • Extended Assay Length – Measure changes in expression over longer time courses than are possible with transient assays.
  • High-throughput Screening – LightSwitch Pathway Reporter cell lines are ideal for both primary and secondary high-throughput screening, and can be used inn many different plate formats (96-, 384- and 1536-well plates).
  • Pathway Profiling – Use a panel of reporter cell lines to characterize the effect of your compounds on a variety of biological pathways.

Custom Services are also available for creating custom stable cell lines or for conducting pathway screening.

LightSwitch Pathway Reporter Stable Cell Lines perform exceptionally in primary screens in 96-, 384- and 1536-well formats, and provide consistent, robust results.

Control 2 Reporter Cell Lines (HT1080 & Hep G2)

The LightSwitch™ Control 2 Reporter Cell Lines are used to normalize for non-specific effects when performing LightSwitch Luciferase Assays using our Pathway Reporter Stable Cell Lines. Each cell line contains a stably integrated LightSwitch ACTB Promoter Control construct (S717678), which is comprised of the promoter sequence of the ACTB (beta actin) gene cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene.

AhR v2 Reporter Cell Line (Hep G2)

The LightSwitch™ AhR v2 Reporter Cell Line (Hep G2) was designed for study of the aryl hydrocarbon receptor (AhR) and the regulation of biological responses to dioxins and toxicity. It contains a stably integrated LightSwitch Promoter Reporter GoClone™ construct (S714555), which is comprised of the promoter sequence of the CYP1A1 (cytochrome P450, family 1, subfamily A, polypeptide 1) gene cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene.

BMP Reporter Cell Line (HT1080)

The LightSwitch™ BMP Reporter Cell Line (HT1080) was designed for study of the Bone Morphogenesis pathway. It contains a stably integrated LightSwitch Synthetic Response Element reporter construct (S900030), which is comprised of repeats of a bone morphogenetic protein (BMP) binding site motif, cloned upstream of a minimal promoter and the RenSP luciferase gene in the LightSwitch Long-range Enhancer Reporter Vector.

GR Reporter Cell Line (HT1080)

The LightSwitch™ GR Reporter Cell Line (HT1080) was designed for study of pathways regulated by the glucocorticoid receptor (GR). It contains a stably integrated LightSwitch Synthetic Response Element reporter construct (S900015), which is comprised of repeats of a glucocorticoid receptor binding site motif, cloned upstream of a minimal promoter and the RenSP luciferase gene in the LightSwitch Long-range Enhancer Reporter Vector.

The LightSwitch™ HIF-1 alpha v1 Reporter Cell Line (HT1080) was designed for study of the Hypoxia pathway. It contains a stably integrated LightSwitch Promoter Reporter GoClone™ construct (S721613), which is comprised of the promoter sequence of the lactate dehydrogenase A (LDHA) gene cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene.

HSF Reporter Cell Line (HT1080)

The LightSwitch™ HSF Reporter Cell Line (HT1080) was designed for study of the Heat Shock pathway. It contains a stably integrated LightSwitch Synthetic Response Element reporter construct (S900017), which is comprised of repeats of a heat shock factor (HSF) binding site motif, cloned upstream of a minimal promoter and the RenSP luciferase gene in the LightSwitch Long-range Enhancer Reporter Vector.

The data at right shows the luciferase activity of the LightSwitch HSF Reporter Cell Line (HT1080) when assayed using the LightSwitch Luciferase Assay Kit following induction by moving the plates to 43°C and then incubating at this elevated temperature for the times indicated (0-8 hours). NFκB v1 Reporter Cell Line (HT1080)

The LightSwitch™ NFκB v1 Reporter Cell Line (HT1080) was designed for study of the Inflammation pathway. It contains a stably integrated LightSwitch Promoter Reporter GoClone™ construct (S721709), which is comprised of the promoter sequence of the IL8 (aka CXCL8, chemokine (C-X-C motif) ligand 8) gene cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene.

p53 Reporter Cell Line (HT1080)

The LightSwitch™ p53 Reporter Cell Line (HT1080) was designed for study of the DNA Damage and Apoptosis pathways. It contains a stably integrated LightSwitch Promoter Reporter GoClone™ construct (S718021), which is comprised of the promoter sequence of the E2F transcription factor 7 (E2F7) gene cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene.

SREBP v1 Reporter Cell Line (Hep G2)

The LightSwitch™ SREBP v1 Reporter Cell Line (Hep G2) was designed for study of the cholesterol biosynthesis pathway. It contains a stably integrated LightSwitch Promoter Reporter GoClone™ construct (S711010), which is comprised of the promoter sequence of the LSS (lanosterol synthase or 2,3-oxidosqualene-lanosterol cyclase) gene cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene. LSS catalyzes the first step in the biosynthesis of cholesterol, steroid hormones, and vitamin D.

SREBP v1 Reporter Cell Line (HT1080)

The LightSwitch™ SREBP v1 Reporter Cell Line (HT1080) was designed for study of the cholesterol biosynthesis pathway. It contains a stably integrated LightSwitch Promoter Reporter GoClone™ construct (S711010), which is comprised of the promoter sequence of the LSS (lanosterol synthase or 2,3-oxidosqualene-lanosterol cyclase) gene cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene. STAT Reporter Cell Line (Hep G2)

The LightSwitch™ STAT Reporter Cell Line (Hep G2) was designed for study of the Interferon pathway and the Signal Transducers and Activator of Transcription (STAT) family. It contains a stably integrated LightSwitch Promoter Reporter GoClone™ construct (S722854), which is comprised of the promoter sequence of the Colony Stimulating Factor 1 (CSF1) gene, which activates STAT1 and STAT3, cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene.
