Each cell line contains a stably integrated human regulatory element, validated to be a biomarker of a specific biological pathway, driving the expression of the optimized RenSP luciferase gene.

LightSwitch Luciferase Pathway Reporter Stable Cell Line Overview

LightSwitch Pathway Reporter Stable Cell Lines simplifies the task of measuring the response of specific pathways to treatment with various compounds or growth conditions. In contrast to transiently transfected reporter vectors, the reporter lines stably express the RenSP reporter gene regulated by an element that is responsive to a specific pathway. Simply seed the cell line in multi-well plates, treat each well as desired, then perform high-throughput luciferase assays to measure the response of the regulatory element to the various treatments.

The AhR HepG2 and AhR HEK293 stable cell lines were assayed for RenSP luciferase following treatment with omeprazole. The HEK293 cell line exhibits a weaker response to omeprazole due to HEK293 cells expressing lower levels of the aryl hydrocarbon receptor (AhR).

LightSwitch Pathway Reporter Stable Cell Lines perform exceptionally in primary screens in 96-, 384- and 1536-well formats, and provide consistent, robust results.

Control 2 Reporter Cell Lines (HT1080 & Hep G2)

The LightSwitch™ Control 2 Reporter Cell Lines are used to normalize for non-specific effects when performing LightSwitch Luciferase Assays using our Pathway Reporter Stable Cell Lines. Each cell line contains a stably integrated LightSwitch ACTB Promoter Control construct (S717678), which is comprised of the promoter sequence of the ACTB (beta actin) gene cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene.

AhR v2 Reporter Cell Line (Hep G2)

The LightSwitch™ AhR v2 Reporter Cell Line (Hep G2) was designed for study of the aryl hydrocarbon receptor (AhR) and the regulation of biological responses to dioxins and toxicity. It contains a stably integrated LightSwitch Promoter Reporter GoClone® construct (S714555), which is comprised of the promoter sequence of the CYP1A1 (cytochrome P450, family 1, subfamily A, polypeptide 1) gene cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene.

BMP Reporter Cell Line (HT1080)

The LightSwitch™ BMP Reporter Cell Line (HT1080) was designed for study of the Bone Morphogenesis pathway. It contains a stably integrated LightSwitch Synthetic Response Element reporter construct (S900030), which is comprised of repeats of a bone morphogenetic protein (BMP) binding site motif, cloned upstream of a minimal promoter and the RenSP luciferase gene in the LightSwitch Long-range Enhancer Reporter Vector.

GR Reporter Cell Line (HT1080)

The LightSwitch™ GR Reporter Cell Line (HT1080) was designed for study of pathways regulated by the glucocorticoid receptor (GR). It contains a stably integrated LightSwitch Synthetic Response Element reporter construct (S900015), which is comprised of repeats of a glucocorticoid receptor binding site motif, cloned upstream of a minimal promoter and the RenSP luciferase gene in the LightSwitch Long-range Enhancer Reporter Vector.

HSF Reporter Cell Line (HT1080)

The LightSwitch™ HSF Reporter Cell Line (HT1080) was designed for study of the Heat Shock pathway. It contains a stably integrated LightSwitch Synthetic Response Element reporter construct (S900017), which is comprised of repeats of a heat shock factor (HSF) binding site motif, cloned upstream of a minimal promoter and the RenSP luciferase gene in the LightSwitch Long-range Enhancer Reporter Vector.

The data at right shows the luciferase activity of the LightSwitch HSF Reporter Cell Line (HT1080) when assayed using the LightSwitch Luciferase Assay Kit following induction by moving the plates to 43°C and then incubating at this elevated temperature for the times indicated (0-8 hours). SREBP v1 Reporter Cell Line (Hep G2)

The LightSwitch™ SREBP v1 Reporter Cell Line (Hep G2) was designed for study of the cholesterol biosynthesis pathway. It contains a stably integrated LightSwitch Promoter Reporter GoClone® construct (S711010), which is comprised of the promoter sequence of the LSS (lanosterol synthase or 2,3-oxidosqualene-lanosterol cyclase) gene cloned into the MCS of the LightSwitch Promoter Reporter Vector upstream of the RenSP luciferase gene. LSS catalyzes the first step in the biosynthesis of cholesterol, steroid hormones, and vitamin D.

The LightSwitch™ SREBP v1 Reporter Cell Line (HT1080) was designed for study of the cholesterol biosynthesis pathway.
