Background
Saos-2 nuclear extract was prepared from cell cultures of the human sarcoma osteogenic Saos-2 cell line. This non-transformed cell line was derived from the primary osteosarcoma of an 11-year old Caucasian female patient. Saos-2 cells exhibit many characteristic osteoblastic properties, including high levels of alkaline phosphatase expression, parathyroid hormone-mediated induction of adenylate cyclase production, and production and secretion of type I collagen and mineralized matrix. Therefore, they are commonly used to study osteoblastic phenotype-related cellular functions and products and in research related to late-stage osteoblastic differentiation.

Saos-2 nuclear extract is specifically recommended for studies related to osteoblasts and osteoblastic differentiation.

Extract Composition
Saos-2 nuclear extract is supplied in 20 mM Hepes pH 7.5, 400 mM NaCl, 20% glycerol, 0.1 mM EDTA, 10 mM NaF, 10 µM Na2MoO4, 1 mM NaVO3, 10 mM PNPP, 10 mM β-glycerophosphate, 1 mM DTT and protease inhibitors. The protein content has been determined by a Bradford-based assay.
Quality Control
Extracts have been quality control tested by Western blot and the Electrophoretic Mobility Shift Assay (EMSA).
Storage
To ensure stability, extracts should be stored at -80°C.
We recommend aliquoting the extracts into single-use fractions and then storing them at -80°C. This eliminates repeated freeze/thaw cycles.
