Validated Applications:
ChIP-Seq: 7 µl (4 µg) per ChIP
WB: 1:500 - 1:1,000 dilution

The addition of 0.05% Tween 20 in the blocking buffer and primary antibody incubation buffer is recommended to aid in detection by Western blot. Individual optimization may be required.

Immunogen

This antibody was raised against a peptide within the N-terminal region of human PBRM1.

Buffer

Purified IgG in PBS with 30% glycerol and 0.035% sodium azide.

PBRM1 antibody (pAb) tested by ChIP-Seq.Chromatin immunoprecipitation (ChIP) was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with 30 µg of chromatin from mouse liver and 4 µg PBRM1 antibody. ChIP DNA was sequenced on the Illumina HiSeq and 3.9 million sequence tags were mapped to identify PBRM1 binding sites. PBRM1 antibody (pAb) tested by Western blot.
The analysis was performed using 40 µg of A-431 nuclear extract (Cat. No. 36004) and PBRM1 (pAb) at a 1:1000 dilution.

Background

PBRM1 (polybromo 1) is a component of the SWI/SNF-B (PBAF) ATP-dependent chromatin-remodeling complex. It is involved in transcriptional activation and repression of select genes by chromatin remodeling and is an integral component of complexes necessary for ligand-dependent transcriptional activation by nuclear hormone receptors. Defects in PBRM1 have been associated with primary clear cell renal cell carcinoma.

Positive Control

Active Motif's A-431 nuclear extract (Catalog No. 36004) can be used as a positive control for PBRM1 antibody.

Chromatin IP Assays

This antibody has been validated for use in ChIP and/or ChIP-Seq, and can be used with Active Motif's ChIP-IT® High Sensitivity Kit or our magnetic bead-based ChIP-IT® Express Kits. For an overview of all of our ChIP products, please visit our Chromatin IP Page to learn about ChIP products designed for use with PBRM1 antibody (pAb).

Storage

Some products may be shipped at room temperature. This will not affect their stability or performance. Upon receipt, unconjugated antibodies may be stored at -20°C for up to 2 years. Fluorophore- & enzyme-conjugated antibodies should be stored at 4°C. Fluorophore-conjugated antibodies should be protected from light. Keep reagents on ice when not in storage; to avoid repeated freeze/thaw cycles, we recommend aliquoting items that will be stored frozen into single-use fractions prior to freezing.
