Applications

ChIP Validated ChIP-Seq Validated Western Blot Validated

Application Notes

Applications Validated by Active Motif:
ChIP: 5 µl per ChIP
ChIP-Seq: 5 µl each
WB: 1:500 - 1:2,000 dilution

ChIP-Seq validation was performed by Active Motif's Epigenetics Services; the complete data set is available in the UCSC Genome Browser by clicking here.

For CTCF, we also offer AbFlex® CTCF Recombinant Antibody (rAb). For details, see Catalog No. 91285.

Published Applications

Immunogen

This antibody was raised against a peptide within the N-terminal region of human BORIS.

Buffer

Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 31 mM glycine, 0.07 mM EDTA, 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

Boris / CTCFL antibody (pAb) tested by ChIP-Seq.
ChIP was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with 30 ug of chromatin from human testis and 2 µl of antibody. ChIP DNA was sequenced on the Illumina HiSeq and 20 million sequence tags were mapped to identify Boris / CTCFL binding sites. The image shows binding across a region of chromosome 7. You can view the complete data set in the UCSC Genome Browser, starting at this specific location, here.

BORIS / CTCFL antibody (pAb) tested by Western blot.
Whole cell extract (10 μg per lane) from HEK293T cells transfected with either vector alone (lanes 1 and 3), with a FLAG-Tagged human BORIS expression vector (lanes 2 and 4) or mouse testis nuclear extract (30 μg, lane 5) was probed with either BORIS / CTCFL antibody at 1:2,000 (lanes 1 and 2) or 1:500 (lane 5) or anti-FLAG-Tag (lanes 3 and 4).

Background

BORIS (Brother of Regulator of Imprinted Sites) / CTCFL (CTCF-like) is a zinc-finger DNA binding protein that acts as a chromatin insulator and a paralog of the CTCF protein. Normally, BORIS / CTCFL is expressed only in primary spermatocytes and is thought to play a role in resetting DNA methylation marks during the differentiation of male germ cells. BORIS / CTCFL is also expressed in many cancers and may serve a pro-proliferative function.

Chromatin IP Assays

This antibody has been validated for use in ChIP and/or ChIP-Seq, and can be used with Active Motif's ChIP-IT® High Sensitivity Kit or our magnetic bead-based ChIP-IT® Express Kits. For an overview of all of our ChIP products, please visit our Chromatin IP Page to learn about ChIP products designed for use with Boris / CTCFL antibody (pAb).
