Note: many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt / Sonication Protocol when preparing nuclear extracts for Western Blot.

Immunogen

This HMGA2 antibody was raised against a peptide within the N-terminal region of human HMGA2 / HMGI-C.

Buffer

Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 31 mM glycine, 0.07 mM EDTA, 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

10 µl of HMGA2 / HMGI-C antibody was used to immunoprecipitate HMGA2 / HMGI-C from 250 µg of HepG2 nuclear cell extract (lane 2). 10 µl of rabbit IgG was used as a negative control (lane 1). The immunoprecipitated protein was detected by Western blotting using the HMGA2 / HMGI-C antibody at a dilution of 1:3,000.

Background

HMGA2 (High-mobility group AT-hook protein 2, HMGI-C) is a member of the HMG (High mobility group) protein family, which are non-histone chromatin-associated proteins involved in DNA organization and regulation of transcription. The HMG protein family consists of six proteins and is subdivided into three subfamilies: the HMG-1/-2 subfamily, the HMG-I/Y subfamily and the HMG-14/-17 subfamily. Mouse models of HMGA2 function indicate that it is involved in proliferation at the cellular level and obesity at the organismal level. Overexpression of HMGA2 is observed in a variety of cancers.

Storage

Some products may be shipped at room temperature. This will not affect their stability or performance. Upon receipt, unconjugated antibodies may be stored at -20°C for up to 2 years. Fluorophore- & enzyme-conjugated antibodies should be stored at 4°C. Fluorophore-conjugated antibodies should be protected from light. Keep reagents on ice when not in storage; to avoid repeated freeze/thaw cycles, we recommend aliquoting items that will be stored frozen into single-use fractions prior to freezing.
