Applications

Western Blot Validated Immunoprecipitation Validated

Application Notes

Validated Applications:
IP: 5 - 10 µl per IP
WB: 1:500 - 1:2,000 dilution

The addition of 0.05% Tween 20 in the blocking buffer and primary antibody incubation buffer is recommended to aid in detection by Western blot. Individual optimization may be required.

Note: many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt / Sonication Protocol when preparing nuclear extracts for Western Blot.

Immunogen

This antibody was raised against a peptide within the C-terminal region of human ATAD2.

Buffer

Purified IgG in PBS with 30% glycerol and 0.035% sodium azide.

10 µl of ATAD2 antibody was used to immunoprecipitate ATAD2 from 250 µg of Jurkat nuclear cell extract (lane 2). 10 µl of rabbit IgG was used as a negative control (lane 1). The immunoprecipitated protein was detected by Western blotting using the ATAD2 antibody at a dilution of 1:1,000.

Background

ATAD2 (ATPase family AAA domain-containing protein 2) may be a transcriptional coactivator of the nuclear receptor ESR1 required to induce the expression of a subset of estradiol target genes, such as CCND1, MYC and E2F1. May play a role in the recruitment or occupancy of CREBBP at some ESR1 target gene promoters. May be required for histone hyperacetylation. Involved in the estrogen-induced cell proliferation and cell cycle progression of breast cancer cells.

Positive Control

Active Motif's Jurkat nuclear extract (Catalog No. 36014) can be used as a positive control for ATAD2 antibody.

Storage

Some products may be shipped at room temperature. This will not affect their stability or performance. Upon receipt, unconjugated antibodies may be stored at -20°C for up to 2 years. Fluorophore- & enzyme-conjugated antibodies should be stored at 4°C. Fluorophore-conjugated antibodies should be protected from light. Keep reagents on ice when not in storage; to avoid repeated freeze/thaw cycles, we recommend aliquoting items that will be stored frozen into single-use fractions prior to freezing.

Guarantee

This product is guaranteed for 6 months from date of receipt.

This product is for research use only and is not for use in diagnostic procedures.
