Background

The SAE1 / SAE2 complex (SAE) is the essential heterodimeric SUMO-activating enzyme that initiates the SUMOylation cascade, a critical post-translational modification regulating diverse cellular processes. SAE catalyzes the ATP-dependent activation of SUMO proteins (SUMO-1, -2, -3) through a two-step reaction: first forming a highenergy SUMO-adenylate intermediate, then transferring activated SUMO to the catalytic cysteine of the E2 conjugating enzyme UBC9 via a thioester bond. This activation step is indispensable for subsequent SUMO conjugation to target proteins by E3 ligases. By enabling SUMO modification, the SAE1 / SAE2 complex governs nuclear transport, genome stability, transcriptional regulation, DNA repair, and stress responses. Dysregulation of SAE function disrupts SUMOylation homeostasis and is implicated in cancer progression (e.g., breast cancer metastasis), neurodegenerative diseases, and viral pathogenesis, making it a potential therapeutic target. Its activity is dynamically regulated by cellular ATP levels and interaction partners like SENP proteases that reverse SUMOylation.

Recombinant SAE1 / SAE2 Complex that includes full length human SAE1 protein (accession number NP_005491.1) without any tag and full length human SAE2 protein (accession number NP_005490.1) with an N-terminal His tag was expressed in E. coli. The molecular weights of SAE1 and SAE2 are 39.83 kDa and 73.39 kDa, respectively.
