Background
SRPK1 (SRSF protein kinase 1) is a serine/arginine-rich protein-specific kinase which specifically phosphorylates its substrates at serine residues located in regions rich in arginine/serine dipeptides, known as RS domains and is involved in the phosphorylation of SR splicing factors and the regulation of splicing. Plays a central role in the regulatory network for splicing, controlling the intranuclear distribution of splicing factors in interphase cells and the reorganization of nuclear speckles during mitosis. Can influence additional steps of mRNA maturation, as well as other cellular activities, such as chromatin reorganization in somatic and sperm cells and cell cycle progression.

Recombinant SRPK1 protein was expressed in a baculovirus expression system as the full-length protein (accession number NP_003128.3) with an N-terminal FLAG tag. The molecular weight of the protein is 75.6 kDa.

ADP-Glo assay for SRPK1 activity
0.1 µg/µl Myelin Basic protein was incubated with different concentrations of SRPK1 protein in a 5 µl
reaction system containing 40 mM Tris-HCl pH 7.4, 20 mM MgCl2, 50 µM DTT, 0.1 mg/ml BSA and 10 µM ATP at 30℃ for 1 hour, then 5 µl ADP-Glo Reagent was added. After 45 minutes incubation at RT, 10 µl Kinase Detection Reagent was added in the system. Following 45 minutes incubation at RT, luminescence was detected with a plate reader.
Storage
Recombinant proteins in solution are temperature sensitive and must be stored at -80°C to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
