Background
MAP3K20 (Mitogen-activated protein kinase 20) is a stress-activated component of a protein kinase signal transduction cascade that promotes programmed cell death in response to various stress, such as ribosomal stress, osmotic shock and ionizing radiation. Acts by catalyzing phosphorylation of MAP kinase kinases, leading to activation of the JNK (MAPK8/JNK1, MAPK9/JNK2 and/or MAPK10/JNK3) and MAP kinase p38. Activates JNK through phosphorylation of MAP2K4/MKK4 and MAP2K7/MKK7, and MAP kinase p38 gamma (MAPK12) via phosphorylation of MAP2K3/MKK3 and MAP2K6/MKK6. Involved in stress associated with adrenergic stimulation: contributes to cardiac decompensation during periods of acute cardiac stress.

Protein Details
Recombinant MAP3K20 protein was expressed in a baculovirus expression system as the full length protein (accession number NP_057737.2) with an N-terminal FLAG tag. The molecular weight of the protein is 92.43 kDa.

ADP-Glo assay for MAP3K20 activity
0.1 µg/µl Myelin Basic protein was incubated with different concentrations of MAP3K20 protein in a 5 µl reaction system containing 40 mM Tris-HCl pH 7.4, 20 mM MgCl2, 50 µM DTT, 0.1 mg/ml BSA and 10 µM ATP at 30℃ for 1 hour, then 5 µl ADP-Glo Reagent was added. After 45 minutes incubation at RT, 10 µl Kinase Detection Reagent was added in the system. Following 45 minutes incubation at RT, luminescence was detected with a plate reader.
Storage
Recombinant proteins in solution are temperature sensitive and must be stored at -80°C to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
