Background
The AHR / HIF1B complex (also termed AHR / ARNT) is a heterodimeric transcription factor that mediates cellular responses to environmental toxins, hypoxia, and metabolic cues. Upon ligand binding (e.g., dioxins or endogenous tryptophan metabolites), cytosolic AHR translocates to the nucleus and dimerizes with its constitutive partner HIF1B (HIF-1β/ARNT). This complex binds xenobiotic response elements (XREs), driving the expression of detoxification enzymes like CYP1A1. Beyond xenobiotic metabolism, it critically regulates immune cell differentiation, particularly in type 1 regulatory T cells (Tr1), where it orchestrates metabolic reprogramming: HIF1α (a HIF1B paralog) initiates early metabolic shifts, while AHR subsequently promotes HIF1α degradation to refine Tr1 cell fate. The complex also modulates macrophage polarization by suppressing glycolysis via the HIF-1α/GLUT1 axis, favoring anti-inflammatory M2 over pro-inflammatory M1 phenotypes. Dysregulation of AHR / HIF1B contributes to inflammatory diseases (e.g., rheumatoid arthritis) and cancer progression, highlighting its role in maintaining immune-metabolic homeostasis.

Protein Details
Recombinant AHR / HIF1B Complex that includes full length human AHR protein (accession number NP_001612.1) with an N-terminal FLAG tag and full length human HIF1B protein (accession number NP_001659.1) with an N-terminal His tag was expressed in Hi5 cells. The molecular weights of AHR and HIF1B are 97.42 kDa and 89.86 kDa, respectively.

1 µM substrate dsDNA (5’ biotin-ATCGTGCGTGACAC 3’) was incubated with different concentrations of AHR / HIF1B Complex in reaction buffer containing 50 mM HEPES-NaOH pH 7.0, 0.1% BSA at room temperature for 1 hour. Anti-FLAG antibody was used to detect reaction products.
