Background

NRAS (GTPase NRas) protein is a critical molecular switch belonging to the RAS family of small GTPases. It plays a fundamental role in regulating cell growth, proliferation, and survival by cycling between an active GTP-bound and an inactive GDP-bound state. Upon activation by upstream signals from cell surface receptors, such as receptor tyrosine kinases, NRAS relays signals through key effector pathways, most notably the RAF-MEK-ERK (MAPK) and PI3K-AKT cascades. These pathways ultimately instruct the nucleus to initiate programs for cell division. Due to its central role in signaling, mutations in the NRAS gene, which lock the protein in a perpetually active GTP-bound state, are frequently associated with uncontrolled cell growth and are found in various cancers, including melanoma and leukemia. Consequently, NRAS is a well-established oncoprotein and a major target in cancer research.
