Background

IFIH1 (Interferon-induced helicase C domain-containing protein 1), also known as MDA5, is a cytoplasmic sensor that is a critical component of the innate immune system. It functions as a pattern recognition receptor (PRR) that specifically detects long double-stranded RNA (dsRNA), a molecular pattern often associated with viral infections. Upon binding to viral RNA, IFIH1 oligomerizes and initiates a signaling cascade that culminates in the production of type I interferons (IFN-α/β) and other pro-inflammatory cytokines, launching a potent antiviral defense state in the cell. Dysregulation of IFIH1 is linked to autoimmune diseases, such as Aicardi-Goutières syndrome and type I diabetes, where its activity becomes inappropriately triggered by self-nucleic acids.
