HTRF assay for MEN1 activity/
3 μM H3K9me2 (1-21 aa) peptide was incubated with different concentrations of MEN1 protein in reaction buffer containing 50 mM HEPES-NaOH pH 7.0, 0.1% BSA at room temperature for 1 hour. Anti-FLAG antibody was used to detect reaction products.

Background

MEN1 (Multiple Endocrine Neoplasia Type 1) is a classic tumor suppressor gene encoding the nuclear protein menin. Menin acts as a critical molecular scaffold that regulates key cellular processes including transcription, genome stability, and cell division. It achieves this by orchestrating the assembly of large protein complexes, most notably with histone-modifying enzymes like the MLL/SET1 histone methyltransferase complex. Loss-of-function mutations in MEN1 disrupt these interactions, leading to dysregulated gene expression, uncontrolled cell proliferation, and ultimately the development of characteristic endocrine and non-endocrine tumors, defining the MEN syndrome.

Recombinant proteins in solution are temperature sensitive and must be stored at -80°C to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
