1 µM substrate dsDNA (5’ biotin-ATATAACTAGGTCATA
3’) was incubated with different concentrations of RORC protein in reaction buffer containing 50 mM HEPES NaOH pH 7.0, 100 µM ZnCl2, 0.1% BSA at room temperature for 1 hour. Anti-FLAG antibody was used to detect reaction products.
Background
RORC (RAR-related Orphan Receptor Gamma) , primarily known as the master transcription factor RORγt, is indispensable for the differentiation and function of immune cells, including Th17 cells and group 3 innate lymphoid cells (ILC3s). It directs the transcriptional program to produce pro-inflammatory cytokines such as IL-17A and IL-22, playing a central role in mucosal immunity, inflammation, and autoimmune pathogenesis. Consequently, RORC/RORγt is a prime therapeutic target for treating autoimmune and inflammatory diseases like psoriasis, multiple sclerosis, and inflammatory bowel disease.
