Background
Recombinant human NAA15 / NAA10 complex, also known as the NatA (N-terminal acetyltransferase A) complex, is a heterodimeric enzyme composed of the NAA15 auxiliary subunit (also called NARG1, NATH, GA19, or Tbdn100) and the NAA15 / NAA10 catalytic subunit (also known as ARD1, hARD1, or TE2). This recombinant complex, typically co-expressed in systems such as E. coli or insect cells, retains its native function as the major eukaryotic N-terminal acetyltransferase, catalyzing the transfer of an acetyl group from acetyl-CoA to the alpha-amino group of nascent polypeptides—a co-translational modification that is essential for protein stability, complex formation, and normal cellular function. Within the complex, NAA15 acts as a scaffold that binds to the ribosome and stabilizes the catalytic NAA10 subunit, while NAA10 performs the enzymatic acetylation of target proteins, including those involved in vascular, hematopoietic, and neuronal growth and development, and regulates key substrates such as HIF1A and TSC2 to influence cellular signaling and homeostasis.
