IP: 2 µg per IP
IF:0.4 - 2 µg/ml
ChIP-Seq:2 µg/ml per IP
Immunogen
This PPARγ antibody was raised against a peptide of human PPARγ.
Buffer
Purified IgG in 140 mM Hepes, pH 7.5, 70 mM NaCl, 32 mM NaOAc, 40% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.
AbFlex® PPARγ rAb antibody tested by Western blot.
BEWO, PC-3, and PANC-1 whole cell extract (80 µg) was probed with AbFlex® PPARγ antibody at 0.2 µg/mL.
AbFlex® PPARγ rAb antibody tested by immunoprecipitation.
2 µg of antibody was used to immunoprecipitate PPARγ from 1 mg of PC3 whole cell extract, 40% IP product (Lane 3) and 40 µg of Input proteins (Lane 1) were then run on SDS-PAGE and probed with the same antibody at 2 µg /ml. Tween-20 at 0.1% was added to blocking buffers to perform Western blot and reduce background. 2 µg of rabbit IgG was used as negative control for IP (Lane 2).
AbFlex® PPARγ rAb antibody tested by immunofluorescence.
PPARγ was detected in human HeLa cells using AbFlex® PPARγ (rAb) at 2 µg/mL, followed by Multi-rAb CoraLite® Plus 594-Goat Anti-Rabbit Recombinant Secondary Antibody (H+L).
AbFlex® PPARγ antibody tested by ChIP-Seq.
Chromatin immunoprecipitation (ChIP) was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with 25 µg of chromatin from PC-3 cells and 2 µg of PPARγ antibody. ChIP DNA was sequenced on the Illumina HiSeq and 18 million sequence tags were mapped to identify PPARγ binding sites. The image shows binding across a region of chormosome 8.
Background
AbFlex® antibodies are recombinant antibodies (rAbs) that have been generated using defined DNA sequences to produce highly specific, reproducible antibodies. Each AbFlex antibody contains a 6xHis Tag, a Biotinylation Tag forenzymatic biotin conjugation using the biotin ligase, BirA, and a sortase recognition motif (LPXTG) to attach a variety of labels directly to the antibody including fluorophores, enzymatic substrates (HRP, AP), peptides, drugs as well as solid supports. AbFlex® PPARγ antibody was expressed as full-length IgG with rabbit immunoglobulin heavy and light chains in mammalian HEK293 cells. PPARγ (Peroxisome Proliferator Activated Receptor Gamma), also called as PPARG or NR1C3, is a member of theperoxisome proliferator-activated receptor (PPAR) subfamily of nuclear receptors. PPARs form heterodimers with retinoid X receptors (RXRs) and these heterodimers regulate transcription of various genes. Three subtypes of PPARs are known: PPAR-alpha, PPAR-delta, and PPAR-gamma.
Chromatin IP Assays
This antibody has been validated for use in ChIP and/or ChIP-Seq, and can be used with Active Motif's ChIP-IT® High Sensitivity Kit or our magnetic bead-based ChIP-IT® Express Kits.
