Applications

Western Blot Validated Immunohistochemistry Validated

Application Notes

Applications Validated by Active Motif:
WB*: 1:500 - 1:1,000 dilution

*Note: many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt / Sonication Protocol when preparing nuclear extracts for Western blot.

Published Applications

The following applications have been published using this antibody. Unless noted above, Active Motif may not have validated the antibody for use in these applications:

Immunogen

This antibody was raised against a peptide within human Smyd3.

Buffer

Purified IgG in PBS with 30% glycerol and 0.035% sodium azide.

Smyd3 antibody (pAb) tested by Western blot.
K-562 nuclear extract (30 µg) probed with Smyd3 antibody at a dilution of 1:500.

Background

Smyd3 (SET and MYND domain containing 3) is a histone methyltransferase. Specifically methylates 'Lys-4' of histone H3, inducing di- and tri-methylation, but not monomethylation. Plays an important role in transcriptional activation as a member of an RNA polymerase complex. Binds DNA containing 5'-CCCTCC-3' or 5'-GAGGGG-3' sequences. Recently, Smyd3 was described as the methyltransferase responsible for a novel site of histone modification, H4K5 methylation,

Positive Control

Active Motif's K-562 nuclear extract (Catalog No. 36015) can be used as a positive control for Smyd3 antibody.
