Background

CDK2 is a serine/threonine-protein kinase involved in the control of the cell cycle, essential for meiosis, but dispensable for mitosis. Activity of CDK2 is maximal during S phase and G2; activated by interaction with cyclin E during the early stages of DNA synthesis to permit G1-S transition and subsequently activated by cyclin A2 (cyclin A1 in germ cells) during the late stages of DNA replication to drive the transition from S phase to mitosis, the G2 phase. Cyclin E/ CDK2 prevents oxidative stress-mediated Ras-induced senescence by phosphorylating MYC. Cyclin E/CDK2-mediated phosphorylation of NPAT at G1-S transition and until prophase stimulates the NPAT-mediated activation of histone gene transcription during S phase.

Application Notes

This product was manufactured as described in Protein Details. Where possible, Active Motif has developed functional or activity assays for recombinant proteins. Additional characterization such as enzyme kinetic activity assays, inhibitor screening or other biological activity assays may not have been performed for every product.  All available data for this product is shown.

Protein Details

Recombinant CDK2 / Cyclin E1 Complex that includes full length human CDK2 protein (accession number NP_001789.2) with a C-terminal His tag and full-length human Cyclin E1 protein (accession number NP_001229.1) with a C-terminal FLAG tag was expressed in Hi5 cells. The molecular weights of CDK2 and Cyclin E1 are 35.57 kDa and 48.77 kDa, respectively.

ADP-Glo assay for CDK2 / Cyclin E1 Complex activity
0.1 µg/µl Histone H1 was incubated with different concentrations of CDK2 / Cyclin E1 Complex protein in a 10 µl reaction system containing 40 mM Tris-HCl pH 7.4, 20 mM MgCl2, 1 mM DTT, 0.1 mg/ml BSA and 20 µM ATP at 30℃ for 1 hour, then 10 µl ADP-Glo Reagent was added. After a 40 minute incubation at RT, 20 µl Kinase Detection Reagent was added to the system. Following 30 minutes incubation at RT, luminescence was detected with a plate reader.

Storage

Recombinant proteins in solution are temperature sensitive and must be stored at -80°C to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
