Background

The Ku70 / Ku80 heterodimeric complex is a crucial DNA damage sensor-scaffold primarily responsible for initiating the Non-Homologous End Joining (NHEJ) pathway, the major mechanism for repairing DNA double-strand breaks (DSBs) in mammalian cells. It rapidly and tightly binds to exposed double-stranded DNA ends, forming a ring-like structure that encircles the DNA duplex. This binding protects the broken ends from degradation and serves as a stable platform to recruit and activate key downstream NHEJ factors. Most critically, Ku loads and activates the DNA-dependent protein kinase catalytic subunit (DNA-PKcs) to form the DNA-PK holoenzyme. The Ku complex then orchestrates the sequential assembly of other essential NHEJ components, including the XRCC4/XLF/DNA Ligase IV complex, facilitating the processing, alignment, and final ligation of the broken DNA ends. Beyond canonical DSB repair, Ku also contributes to maintaining telomere stability and is essential for V(D)J recombination during immune system development, thereby playing a fundamental role in preserving genomic integrity.

Protein Details

Recombinant Ku70 / Ku80 Complex that includes full length human Ku70 protein (accession number NP_001275905.1) with an N-terminal His tag and full length human Ku80 protein (accession number NP_066964.1) with an N-terminal FLAG tag was expressed in Hi5 cells. The molecular weights of Ku70 and Ku80 are 72.94 kDa and 83.97 kDa, respectively.
