Immunogen
This Ku80 antibody was raised against a peptide of human Ku80.
Buffer
Purified IgG in 140 mM Hepes, pH 7.5, 70 mM NaCl, 32 mM NaOAc, 40% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.
AbFlex® XRCC5 (Ku80) rAb antibody tested by Western blot.
20 µg of whole cell extracts were run on SDS-PAGE and probed with AbFlex Ku80 antibody at 0.5 µg/ml.

Immunoprecipitation assay of AbFlex® XRCC5 (Ku80) antibody
(rAb)
2 µg of antibody was used to immunoprecipitate Ku80 from 1 mg of HEK293 whole cell extract, 40% IP product and 10% Input were then run on SDS-PAGE and probed with the same antibody at 0.2 µg/ml. Tween-20 at 0.1% was added to blocking buffers to perform Western Blot and reduce background. 2 ug of rabbit IgG was used as negative control for IP.
Immunofluorescence stain of AbFlex® XRCC5 (Ku80) antibody (rAb)
Ku80 was detected in human HeLa cells using AbFlex Ku80 (rAb), followed by Multi-rAb CoraLite® Plus 594-Goat Anti-Rabbit Recombinant Secondary Antibody (H+L).
Background
AbFlex® antibodies are recombinant antibodies (rAbs) that have been generated using defined DNA sequences to produce highly specific, reproducible antibodies. Each AbFlex antibody contains a 6xHis Tag, a Biotinylation Tag forenzymatic biotin conjugation using the biotin ligase, BirA, and a sortase recognition motif (LPXTG) to attach a variety of labels directly to the antibody including fluorophores, enzymatic substrates (HRP, AP), peptides, drugs as well as solid supports.
AbFlex® Ku80 antibody was expressed as full-length IgG with rabbit immunoglobulin heavy and light chains in mammalian HEK293 cells.
XRCC5 (X-ray repair cross complementing 5, also known as Ku80, KUB2, Ku86, NFIV, KARP1, KARP-1) is the 80-kilodalton subunit of the Ku heterodimer protein which is also known as ATP dependant DNA helicase II or DNA repair protein XRCC5. Ku is the DNA-binding component of the DNA-dependent protein kinase, and it functions together with the DNA ligase IV-XRCC4 complex in the repair of DNA double-strand break by non-homologous end joining and the completion of V(D)J recombination events.
