Background

The HIF1A / HIF1B complex serves as the master regulator of cellular adaptation to hypoxia, where oxygen deprivation stabilizes HIF1A, enabling its dimerization with constitutively expressed HIF1B to form the active HIF-1 transcription factor. This complex binds to hypoxia-response elements (HREs) in DNA, activating hundreds of genes critical for maintaining oxygen homeostasis. Key functions include promoting angiogenesis (via VEGF), enhancing glycolytic metabolism (through GLUT1 and glycolytic enzymes), stimulating red blood cell production (via EPO), and supporting cell survival under low-oxygen stress. Beyond physiological adaptation, HIF-1 drives tumor progression by helping cancer cells thrive in hypoxic microenvironments, facilitating metastasis, and conferring therapy resistance. It also modulates immune responses by influencing macrophage polarization and T-cell differentiation. Dysregulation of this complex contributes significantly to ischemic diseases (e.g., stroke, heart attack), inflammatory disorders, and over 70% of human cancers, positioning it as a major therapeutic target across multiple disease contexts.

Protein Details

Recombinant HIF1A / HIF1B Complex that includes full length human HIF1A protein (accession number NP_001521.1) with an N-terminal FLAG tag and full length human HIF1B protein (accession number NP_001659.1) with an N-terminal His tag was expressed in Hi5 cells. The molecular weights of HIF1A and HIF1B are 93.94 kDa and 89.86 kDa, respectively.

HTRF Assay for Recombinant HIF1A / HIF1B Complex activity
1 µM substrate dsDNA (5’ biotin-AGTCGCACGTGCGAAT 3’) was incubated with different concentrations of HIF1A / HIF1B Complex protein in reaction buffer containing 50 mM HEPES-NaOH pH 7.0, 0.1% BSA at room temperature for 1 hour . Anti-FLAG antibody was used to detect reaction products.

HTRF Assay for Recombinant HIF1A / HIF1B Complex activity
1 µM substrate dsDNA (5’ biotin-AGTCGCACGTGCGAAT 3’) was incubated with different concentrations of HIF1A / HIF1B Complex protein in reaction buffer containing 50 mM HEPES-NaOH pH 7.0, 0.1% BSA at room temperature for 1 hour . Anti-FLAG antibody was used to detect reaction products.

Storage

Recombinant proteins in solution are temperature sensitive and must be stored at -80°C to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
