Background

POLQ, also known as DNA polymerase theta, is a unique and specialized DNA polymerase that plays a critical role in an alternative, error-prone pathway for repairing double-strand breaks, known as theta-mediated end joining (TMEJ). Unlike classical repair mechanisms, TMEJ functions as a backup system to ligate broken DNA ends with minimal homology, often resulting in mutations and deletions. While this promotes genomic instability, it is essential for the survival of cancer cells that have defects in other repair pathways like homologous recombination. Consequently, POLQ is overexpressed in many cancers and has emerged a promising therapeutic target. Its functions also include mitigating replication stress and protecting against DNA end resection.
