Background

NR1D2 (Nuclear Receptor Subfamily 1 Group D Member 2), also known as Rev-Erbβ, is a transcriptional repressor and a core component of the circadian clock machinery. It functions as a heme-binding nuclear receptor to rhythmically repress the expression of key clock genes, such as BMAL1 (ARNTL), thereby regulating circadian physiology. Beyond timekeeping, NR1D2 integrates metabolic and inflammatory pathways, influencing lipid and glucose homeostasis, as well as macrophage function. Dysregulation of NR1D2 is linked to circadian rhythm disorders, metabolic diseases, and inflammatory conditions.
