HTRF Assay for Recombinant NR2C1 activity.
1 µM substrate dsDNA (5’ biotin-ATAGGTCACGGAGAGGTCAGC 3’) was incubated with different concentrations of NR2C1 protein in reaction buffer containing 50 mM HEPES-NaOH pH 7.0, 100 µM ZnCl2, 0.1% BSA at room temperature for 1 hour. Anti-FLAG antibody was used to detect reaction products.

Background

NR2C1 (Nuclear Receptor Subfamily 2 Group C Member 1)
, also known as TR2 (Testicular Receptor 2), is a nuclear receptor that functions as a transcriptional repressor or activator in a context-dependent manner. It is involved in embryonic development, cell proliferation, and differentiation, particularly within the hematopoietic and nervous systems, by regulating gene networks related to growth and metabolism. Emerging research also links its dysregulated expression to the development and progression of certain cancers.
