Background

Histone H1.X, also known by its gene symbol H1FX or alternative names such as H1-10, H1.10, and H1 histone family member X, is a replication-independent subtype of the linker histone H1 family that exhibits distinct biochemical properties and plays specialized roles in chromatin dynamics. Unlike the more abundant somatic H1 variants, H1.X demonstrates significantly lower DNA, nucleosome, and histone chaperone binding activities, which results in its remarkably rapid exchange on and off chromatin compared to other H1 subtypes. This highly dynamic linker histone accumulates in the nucleolus during interphase and localizes to the chromosome periphery during mitosis, where it has been shown to be essential for proper chromosome alignment and segregation. Furthermore, H1.X displays unique chromosomal binding site preferences that differ from other variants like H1.0, with its N-terminal, globular, and C-terminal domains cooperatively determining its specific genomic localization patterns and contributing to its regulatory functions in higher-order chromatin structure.
