ELISA for PARP14 (737-1801) activity
25 μM NAD-Biotin was added into ELISA plate (coated with histone H2A and H2B mixture) with PARP14 (737-1801) in ADPR Buffer and incubated for 1 hour at room temperature. Following Streptavidin-HRP and ECL incubation, the plate was read in a luminometer or microtiter-plate reader.

Background

Recombinant human PARP14 (poly(ADP-ribose) polymerase 14), also known by its gene symbol PARP14 (poly(ADP-ribose) polymerase 14) and alternative names including ARTD8 (diphtheria toxin-like ADP-ribosyltransferase 8), BAL2 (B-aggressive lymphoma 2), and COAT6, is a mono-ADP-ribosyltransferase that catalyzes the transfer of ADP-ribose from NAD+ to target proteins, thereby regulating diverse cellular processes including DNA repair, transcription, and cell signaling. This recombinant form, typically produced in expression systems such as E. coli or insect cells, retains the key structural and biochemical properties of the native protein, including its macrodomains that bind ADP-ribose and its WWE domain involved in protein-protein interactions, along with the catalytic PARP domain responsible for mono-ADP-ribosylation activity. PARP14 (poly(ADP-ribose) polymerase 14) functions primarily as a pro-survival factor by promoting STAT6-dependent gene expression, inhibiting apoptosis through interactions with the JNK signaling pathway, and serving as a key regulator of B cell differentiation and macrophage activation, while also contributing to the DNA damage response and playing context-dependent roles in cancer progression and immune cell function.
