Immunogen

This EGLN2 (PHD1) antibody was raised against a peptide of human EGLN2 (PHD1).

Buffer

Purified IgG in 140 mM Hepes, pH 7.5, 70 mM NaCl, 32 mM NaOAc, 40% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

U2OS, HeLa and A549 whole cell extracts (40 µg) were probed with AbFlex® EGLN2 (PHD1) antibody at 0.2 µg/mL.

Background

AbFlex® antibodies are recombinant antibodies (rAbs) that have been generated using defined DNA sequences to produce highly specific, reproducible antibodies. Each AbFlex antibody contains a 6xHis Tag, a Biotinylation Tag forenzymatic biotin conjugation using the biotin ligase, BirA, and a sortase recognition motif (LPXTG) to attach a variety of labels directly to the antibody including fluorophores, enzymatic substrates (HRP, AP), peptides, drugs as well as solid supports. AbFlex® EGLN2 (PHD1) antibody was expressed as full-length IgG with rabbit immunoglobulin heavy and light chains in mammalian HEK293 cells. Prolyl Hydroxylase Domain Containing Protein 1 (PHD1) or Egl-9 Family Hypoxia Inducible Factor 2 (EGLN2), also known as HIF-PH1 or EIT6, is a prolyl hydroxylase. PHD1 is a cellular oxygen sensor that catalyzes the post translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins under normoxic conditions. It can hydroxylate a specific proline found in each of the oxygen-dependent degradation (ODD) domains (N-terminal, NODD, and C-terminal, CODD) of HIF1A, also HIF2A. PHD1 prefers the CODD site for both HIF1A and HIF2A.NODD, and C-terminal, CODD) of HIF1A, also HIF2A.
